To test the hypothesis of Syndrome and Treatment Pharmacokinetics (S-TPK)
the authors have studied the pharmacokinetics of tetramethyrazine phosphate (TMPP) in healthy and blood stasis dogs by high performance liquid chromatography. After a single intravenous injection of TMPP 8 mg kg-1 to healthy and blood stasis dogs
compartment model of TMPP serum concentration was fitted and then pharmacokinetic parameters were calculated with a Microcomputer Pharmacokinetic Program on a Compaq 80386 computer. Important parameters were as follows: in the healthy dogs
T1/2P = 30. 27 ± 6. 85 min
Kel = 0.11 ± 0. 04 min-1
AUC= 445.26±51.26 mg/L min; in the blood stasis dogs
T1/2β = 71.58β10.80 min
Kel = 0.06 β0.01 min-1
AUC = 1381.85β464.68 mg/L min. Through this experiment it was found that there was significant difference in the pharmacokinetic parameters between healthy and blood stasis dogs. The results obtained are consistent with S-TPK hypothesis.
