摘要

Objective: To explore the inhibitory pathway and the possible mechanism of panicoin on platelet activation.Methods: The effect of panicoin on platelet aggregation

thromboxane B2 release

P-selectin (GMP-140) count

levels of cAMP and cGMP induced by ADP

epinephrine

arachidonic acid or thrombin were observed.Results: Panicoin could inhibit significantly platelet aggregation induced by ADP

arachidonic acid and epinephrine in a dose-dependent manner

thrombin induced GMP-140 on platelet surface and TXB2 release

and increase the platelet cAMP level

but showed no effect on cGMP level.Conclusion: Panicoin shows a potent inhibition on platelet activation and antithrombotic effect. The mechanisms seems to be related with elevation of platelet cAMP.
